What should be the difference in content between title tags and description tags?

Title tag length should be about 65 characters and it should contain the best keywords related to page. Where as length of description tag should be about 255 characters and it should contain description of page or you can say description of title.

I have seen many site on top of google whhich are missing description tag but containing title tag containing specific keyword.

That is what I just said. Description tags are more important for visitors to see what the site is about. By using the right description, you can optimize the CTR.

That depends. If you want to do brand marketing, yes. However, if you just started out and are not really a brand, it makes no sense and will not be beneficial.
