As long as you're logged into your account, If you go to their twitter account you should have an option to unfollow. It should be as simple as that. Unless their is something else specifically you're trying to do.

I use ManageFlitter for unfollowing a large amount of people quickly. It's free and will only unfollow those who don't follow you back.

Do not, and I repeat, do not mass unfollow anyone, especially via a 3rd party site. Not if you value your twitter account. That's just lousy advice and completely against the TOS of twitter. Ditto with following people. The right to do it is with a trickle. Go to friendorfollow.com (notice I'm not giving a link? No reason to.) be logged into twitter and enter your handle. it will show you all the people who you follow that don't follow you back. Click on each one, manually, and unfollow roughly 10 a day.
