I'm going to start drop shipping with Shopify, what is a good estimate of the start up cost that's involved and some good wholesalers you have used.

I'm afraid that not many people will be able to help you with the (lack of) information you've provided.

I know you're planning to drop ship using Spotify as a platform. So, you're going to set up an ecommerce store on Spotify.

Are you going to design this shop and upload the products yourself? What, if anything, are you capable of doing? Are you going to outsource anything or everything? The more you do, the less your cost will be.

Depending on your requirements, you could outsource the work on oDesk or Elance anywhere between a couple hundred dollars and a couple thousand dollars. It really depends on your requirements. How many products? Are they provided via a feed, or will they need to be added individually?

The biggest hurdle in drop shipping, in my opinion, is finding reliable drop shippers and products with high enough profit margins that it's worth selling. In my experience, the profit margins are slim, unless you are able to set up a unique deal with a manufacturer/supplier. Most of the generic "we drop ship" sites offer to drop ship for just about anyone. Meaning lots of competition and lower profit margins. Those who have set up unique and profitable deals with manufacturers will never reveal their sources, unfortunately.

If you do it all yourself, literally less than $100 to start. I am not exaggerating. Start on their lowest plan at 29.99/month, use Shopify payments and miscellaneous fees for things like licensing.

As for wholesalers, very few people will share their best wholesalers with you. I know I wouldn't as I would not want you to compete with me.

As was said above, take the time to really figure out your niche, what you want to sell, where there is a need and if you can buy with at least a 30% gross margin (minimum). Once you have done that, getting started is easy.
