Help: Posting first article - Advice needed

So, I've chosen a niche, chosen 3 keyword phrases, done the research and feel that I have a good chance of ranking well for the keywords.
and of course I also have small detail questions :confused:
any help will be greatly appreciated
I'm publishing an ezine article as I heard it ranks well and are the best for click through to my affiliate's site.
so first question
what should I do for onpage optimization of my article? how much to include the keyword (keywords?) in the article?
The article title will become the ezine domain extension, so should I include the main keyword in the title?
if my keywords were "increase height" "increase your height" and "naturally increase height"
would a title that "best 5 ways to naturally increase your height" be a good title for all three keywords or would the keyword "increase height" not be optimized for since that specific phrase is not in the title?
basically would having the words spread apart in the domain still count as being optimized for that phrase?
(this is not the niche by the way)
second question is about ezine. what is ok to put in the bio box to get the best CTR but not get my article removed. what's considered kosher with ezine?
also, if for one of my keywords there already is an ezine article ranking up there and probably has been there for a while, did google develop any preference for it, or will it replace it with my article if I do better SEO otherwise?
and lastly should I also submit this article to other directories? which ones? Remember this is not about submitting it to as many places as possible to get links, it's about ranking to get traffic and make affiliate sales.
