All in one SEO plugin for Wordpress

I currently have the all in one SEO plugin on my blog and have the following question. When on the add new post page I scroll down to the bottom and see the all in one SEO pack box. What should I be typing in this box if anything before I publish my new post. Do I fill in this box everytime I make a post or is it something that i should fill in only once.
I hope this makes sense.

      I am currently using a program that automatically inserts videos to my blog, so sometimes do not make manual posts. Do i fill in the SEO box with the video title every time one gets placed onto my blog
