This is a small online company that has a growing social network. I'm being interviewed for a sales position in which I would prospect for businesses to advertise on their website. The recruiter told me to be ready to talk about how I would prospect for new business, as they target any large advertiser outside of the Fortune 500.

Any suggestions how I could best answer this question?

I think you would need to understand how your service would benefit an advertiser before you approach them, or their advertising agency, more likely. You would need to look at the company's website, make further inquiries about them if you need to, figure out with the help of your employer what it is you have to offer them that is of benefit to THEM (not you), and then put together an offer.
