When optimizing an Adwords campaign from scratch is there a specific order in which variables should be optimized for maximum ROI?

ie) Keywords, then GEO, then days of Week, then Hour of day?

Or is there another way to do it?

I recommend you to split test your ads.
Try different keywords with different snippets. Then check your stats. Try to optimize the ad that worked best and again try to split test with small changes.
This is how I optimize my ads.

The most important factor can be the keywords targeting. The easiest way to find "hot' keywords for your campaign is to look at competition in your niche that is doing good or killing it in the niche, look at what they are going after. This can save you tons and tons of cash and time.

Then move on to ad (constant testing) and so on.

If only dealing in certain areas, then automatically use geo targeting and if nationwide can go after nationwide but also suggest testing certain areas as well with geo targeting so can find hot spots.

As far as hours and days ad runs, i always suggest running with your business hours unless selling 24 hours a day. If you are open m-f 9 to 5 then run ad for that so you can take the call. If selling a prodcut or generating leads, then can always run all day 7 days a week, however you may find wated times as well, so if running 24 hours a day and see all clicks after 3am -9am are junk, then could eliminate after 3am-9am and etc....

You want to test all aspects of campaign, but again, all starts with the keywords.

Sounds good, but how can you tell if a competitor is doing good or killing it in the niche?

      What criterion do you use?

We do not have access to a competitors statistics. So we really cannot tell if what they are doing is working or not working. If the competitor has been doing the same thing for a long time, it would signal that whatever they were doing was working. If it wasn't working, they would not keep doing it. However, I do not know of any source for historical data that allows us to look back and see if the competitor has been doing the same thing for a long time.

So how can you tell if a competitor is really doing well?
