I've just created a new site and was hoping for a little guidance from all you SEO specialists out there.

Let's say the domain is - keyword-brand.com (title tag: Keyword Brand)

Then I have a first level page - keyword-brand.com/subject-one.html

Is it best to have the title tag for that page as:

Subject One

or

Keyword Brand | Subject One

Thanks in advance

    Keyword Brand Subject One

...and you should name your page keyword-brand.com/keywordbrand.html or if you are going to have several subject pages
    keyword-brand.com/keywordbrandsubjectone.html
    or keyword-brand.com/keywordbrand_subjectone.html

This is how you get a nice letter from a lawyer representing the brand that tells you to cease and desist.

But, to get to your question, I've used both methods on a number of sites and I haven't seen it have a major impact one way or another. My preference has become to only use the page title since this tends to work better with certain tools I use.
