There is some debate over whether a subdomain is treated as a separate domain, I don't believe it to be the case in these kinds of situations, which would lead me to suggest that either is equally good in terms of search engines. But if you want to be on the safe side go with site.com/blog

    It's not an either/or question.

It depends on on a whole bunch of factors. Generally, if mysite already has content, is indexed and ranking and the blog is directly related to the main site, it's better to create a subfolder than create a separate site on a sub-domain.

I would go for the "mysite/blog" option. I am not a huge expert, but the fact is that you don't see a lot of results from the other option on the #1 page of google

Subdomain is typically treated differently, unless you make it a copy cat theme of the main page.

Same here. I would go for "mysite/blog" option. I find this option visitor-friendly.

I think that first one is better then second because if you use blog.mysite.com then google consider it different domain and this become referral domain to your main website. That is not done in mysite.com/blog url because google consider it as a sub folder of a website. You can see this thing in your website's analytic report. That's why second one is good to use.

I think subdomains are always separate domains. The easiest example can be given to you is, if you get a subdomain from blogger or wordpress. They are treated as separate domains without any exception.

If you go with the first one then you can pass some good link juice to your blog and it helps in SERPs.

If you go with blog.mysite.com. Google treats it as a different site.

See it this way..... xyz.blogspot.com and abc.blogspot.com...
you need to work on both of them to get them ranked even when blogspot.com is a authority site...

If you are already running a site on mysite.com, it would be better to make a blog on mysite.com/blog.

I'm personally not a big fan on sub-domains, but apart from that, what UMS (Peter) said above...

If you use a subdomain then it is like starting a new domain from scratch and the good thing about this is that if there is a penalty from search engines then only the subdomain is affected.

Eg: Hubpages.com changed their site url from subfolder to subdomain after google panda update.
