It really depends on your niche and general your readership and blog, if it's the blog on your signature I would suggest to add a leaderboard ad banner below the navigation menu, it's one of best places for adsense for me, however have in mind that niches like blogging , IM, technology don't go very well with adsense since people are very used on that kind of ads.

Anyway I have published a few posts about the best use of adsense on my blog and I am sure that you will find them helpful, take a look below:

You can try an adsense optimized theme like the CTR Theme, (it's what I use) it can rotate through the different layouts automatically and if you use adsense channels you can find your best CTR on a particular layout. (then make it permanent)

I am using eleven40 Genesis child theme because of its responsive design and load time. Maybe i shoud look for a plugin that can rotate the ads on the theme.

Yes, you should try the CTR theme, I get an average 37% CTR, and it loads very fast (.69 seconds) But I also use cache plugins to increase load times

The CTR theme is not responsive, but it displays well on mobile sites. I wouldn't think a responsive theme would help your CTR (earnings) either.

Did you really mean to say 37% CTR? Is that not a massive outlier to the norm for Adsense?

It wont of course but i've always been a fan of responsive designs cos the benefits are awesome. All the same i will look for a CTR theme and give it a test.

While a theme can help you it's impossible to give you good advice without more info.

What's your traffic? What's your CTR? Your niche?

Those are more important things to know. No theme is going to magically make you money.

Simple page with a single text Ad block top/center, do that for a couple of weeks. Assumes you won't mess with the Ad position for a couple of weeks, have niche traffic on the page, & not have a page bloated with distracting bright colored images.

Build more backlinks to your blog. This will help it to rank higher in the search engines, get more organic traffic, and ultimately earn more money.

Also, you may want to change the color scheme of your ads, to make them look like they're a natural part of the page. Positioning of the ads is also important. Ads that are at the top or to the left of the page get more clicks.
