You just need to start testing. That's the only way you can really ever know for sure. I could tell you to use a specific layout, but what works for one site, may not work for another, it's really dependent on a LOT of different variables.

    For example:

    *overall niche
*type of keyword visitor used to come to your site
    *placement of ad
    *colour of title, description, URL

    To name a few.

Does your site get much traffic? basically you want to use a decent number of page views to test each variable. I recommend starting off with the ad placement, as I think that's probably the highest leverage testing you can do to begin with.

Choose one type of placement (or arrangement of ads) keep notes on how you have it setup, and when you implemented it. Then let it sit for a good number of page views to get your data. I would say you'd want to test with AT LEAST 200 page views when testing adsense responses, but that's up to you.

Once you have your data (CTR, CPC - whatever else you think's important) from your chosen amount of pageviews for testing, note it down with your other information (a spreadsheet would be the best idea) and then choose another layout. Do this until you are satisfied you've tried enough different types of layout, and then you'll have your best performer.

Then it's just a matter of moving onto the different types of changes (colours, fonts, etc) and running those changes through the same process.

It's all about testing. Really conversion and optimizing testing should be an ongoing process, but that depends how diligent you are

    Hope it helps!

you can either test using all the information I just wrote up for you, or you can change your niche and start over.

CPC depends from which country you get the visitors. If they are from US or Canada, you will have bigger CPC.

Unfortunately, those cpc figures you see in the adwords is heavily over-estimated. Use google's contextual tool.

Ive seen keywords valuing at $50+ per click in adwords keyword tool, but inside the contextual tool, it shows the harsh reality of $1-2 per click.

Choose your keywords from the contextual tool's figures.
