Of lately I am seeing these types of search queries for my site: www.mydomain/wp-login.php.

Does this mean that someone is trying to hack my site? As of writing, only today there are over 100 such queries for my site. They are coming from different IP addresses. Some from Russia, Brazil, Thailand etc.

What measures should I take to combat this barrage? Why are people doing this?

    Change you WP login url to something random.

    you can try "limit login attempts " plugin of wordpress ,It will do the job
