Do upper and lower place characters play a part in Keywords and SERP's?

I was just trying one of those online SERP checkers on a site I made and I typed in the keyword i wanted to check SERP for, it came back not placed in top 50. I thought that was weird as I was at position 16 the day before, so I tried it again, and got the same not it top 50. Thinking maybe I was spelling it wrong, I copied and pasted the keyword from my header tag and put that in the search field instead, but this time it came back as pos 16 again.
The only difference is that when I type the keyword in with uppercase first letters (like in my header tag) it is positioned higher in SERP.
What's that all about? Quite surprised as I didn't think capitalisation mattered in SEO.
