I wouldn't concern myself with no-follow do-follow any more. Google has so discounted blog commenting backlinks that they're not worth if (for SEO value that is).

However, making intelligent comments on authority blogs related to your niche can result in REAL, Immediate, targeted traffic. Just try to add value to the post.

If the links are nofollow, they're nofollow. In my opinion they're not totally worthless, but I agree with DavidOlsen. You shouldn't comment just for the backlink.

"Dofollow blog lists" is someone saying "hey, try to post spam here". I don't think they're a good way to get backlinks.

Links in signature will not help your SEO. Not on this site, and not on any other forum.

Sure there's worth. Relevant comments on relevant blogs can make a difference. As long as there arent hundreds or thousands of comments.

Adding a quality comment on a top industry related blog is a really good way of generating exposure as it shows people that you are an expert in your niche. Don't worry yourself about 'nofollow' or 'dofollow' and think about the purpose of your comment. If posted on top quality sites, they are all good links regardless.

That depends entirely on the site, and the level of visitor engagement that the site enjoys. People are there to read the piece, not the comments. Unless the comment system is built to highlight the very best replies, they're probably just sinking in the mass. In addition, it takes more than a casual observer to judge anyone's expertise from comments on someone else's blog.

Nofollow links are nofollow. They will show in your site's link profile, but by definition don't add google juice to it. Blog posts are also going to drop from the front page of an active site in a few days, and it's likely that they don't have much link power in any case.
