Your keywords have to be used throughout your site copy - where they make sense - as well as in your title tag and other meta tags. Don't go nuts and start inserting keywords every other word, because this is look at bad form.

I'm quite confused as well, are you referring to the keywords themselves or the tool to get a hold of popular keywords?

Select the keywords that you can compete with easily and fast at start. It would be better if your keyword is your domain name. Once you're up, move to high competitive keywords then.

Before you start building your website, you should already know your keywords. So, hopefully you haven't started building your website yet or you're probably in for much more work than originally thought.

Type a search in google for your specific keywords that you're considering (that revolve around your topic) and see how many results pop up. Now consider a similar keyword phrase and search it.. How many results show for this? The goal is to have a long tail keyword phrase that accurately depicts your content, with very little competition showing for this keyword phrase.

Now, you should always put these searches and information into a database (i.e. excel) This will make it easy to browse and determine what to target. Then, take all of these phrases (starting with the least competition) and enter them into a keyword tool like Google Keyword tool. Find the one with the highest (exact) searches per month, (local). You'll want to put most of your emphasis on these.
