How would it effect rankings if we use two different anchor texts for same domain in one article or in profile or in forum signature?

will ranking go high for both anchor texts or for one? or would have negative effect?

I recommend using a few different anchor variations, all containing your main key phrase. This has the effect of making the links appear more 'natural'.

2.Google always considers the first anchor text to a link. However we can change it by re-ordering the site architecture.

Why not link from one anchor text to homepage and another one to one of your internal page?
The point is the anchor text should be targeted to a page which is particularly optimized for that keyword.

        Hi,

I use usually three search terms in the Bio Box, which all are related to the site theme ( of course ).

I hardly never use the same keyword, which I have used for the site on-page
        optimization.

The important thing is, that the keyword tells to the search engine and to the reader, what is the theme and the benefit a reader can get by visiting the landing page.
