Check the actual post your page appears on to see if it's indexed in Google. If it is, Google will know of the link's existence. If the page doesn't exist, then Google won't know of its existence.

Yes! What carries the most weight is the PR of the Root domain.

In both cases you can continue with building links as your link will be placed on a new page, so no need worry about.

Keep continue building links.This helps in getting more links even some links missing we still manage to get backlinks.Always use pingers and rss submission.
