Keyword research do's and don'ts

I am in Canada. A great keyword IS available as a .ca but .com .net, .org are taken. Do I take the .ca and build a site or do I keep looking for a .com .net .org and use a stop word/letter? If I put "my" in front of the keyword (or something like that) to get a .com .net .org is that better than using and registering the .ca?

If I have an indexed website already that is not related to the keyword or is partially related should I optimize the new keyword and post on that existing domain.

Example: symptomsforlymesdisease.com/symptomsforheartburn

The "symptomsfor" being in the domain and the post or do I have to stick with "lymesdisease" information?

Thank you.

If you are only targeting Canadians, you might as well get .ca. If you are just trying to rank in the US or worldwide get one of the other 3 com/net/org, with whatever addition you want (I usually add something at the end).

      Like tips at the end?

What about the existing site symptomsforlymesdisease.com/symptomsforheartdisease

"symptoms for heart" disease being the keyword for the post.

Or do I need to get another domain symptomsforheartdisease.com

      Thank you
