Just curious what the best Tier 1 links to use are.

I've read 2.0's (created as if they were your money site), Articles, Press releases, High PR links on relative blogs or sites. Am I missing any?

Also should the first Tier all be built manually or can you make them using like GSA for example?

Yes I do agree with you. Start with contextual links using unique articles is better. (Web 2.0 blogs, press releases and etc) - Do not forget to promote those links in good way as well.....

Quality content that is unique around your T1's is best too. It doesn't have to be 100% of the time for all them but it's really is best.

What you can also do is to check all backlinks pointing to sites of your competitors that already rank well and analyze them (using SEO factors that determine value of this or that backlink). And when you have a few backlink profiles uncovered, it's easier to see the pattern and pick the backlink sources for Tier 1.

By the way, the WebMeUp backlink checker is completely free as for now and doesn't even require any specific registration - http://webmeup.com/tools/backlinks.htmlhttp://webmeup.com/tools/backlinks.html

What will you do with all your zero authority web 2.0 links, are you collecting them just because you can?

I personally prefer Magic Submitter / Money Robot Submitter's Web 2.0 and other high PR websites for Tier1 and then i use Article Kevo / GSA SER for tier2 links
