You need some content in there to get google to think it's relevant to your keyword. You don't want to be taken to a site that is completely unrelated just because the backlink profile is good.

Personally I don't really build tiered links. Why should it pass link juice, if you're just spamming them anyway? Just make sure your tier 1s are indexed.

Make sure your video is fully optimized. This means add tags and everything as well as making the title SEO and user friendly. After that there's really just creating quality links to the video.
