Duplicate content is the biggest thing. Crappy backlinking with crappy half spun content might work for a few weeks but it is not strong enough to last.

If you mean on-page SEO, then yes it is very important to have the right tags (title, meta, keywords, h1, h2, etc.) but I would think you'd still need some backlinks to beat a PR3 or PR4 that already...

Multiple backlinks from all the pages on a wesite is not going to get you as much juice as you think. I'd rather one in content link on the highest PR page (usually home page)
