I've got 3 ebooks on Amazon, one of which is selling quite well. However, I simply create the book myself as a .doc and then upload it directly on the DTP site. Admittedly, my ebooks are only 25-40...

Amazon is a good starter. From there you'll want to publish with Smashwords who will push it out to multiple formats and distributors, including Barnes and Noble, Sony, Apple, etc.

One other comments for those unaware:

Submit to Smashwords. It takes a little more time but Smashwords will push it to Barnes and Noble, Sony, Apple, etc. They will create it in pdf, etc.
