I'm still learning at this as well but what is working for me at the moment is to use tweepi to unfollow people that are not following you back the find other people or companies in your same niche then use tweepi to follow their followers! Follow 100 people a day for two days then unfollow peolpe not following you on the third day! repeate this and you will find your twitter followers will grow! obviously you have to tweet good content to keep your followers interested. Avoid buying the follwers as they won't be real persons obviously, Follow the people within your niche, you'll get some of them follow you back, use the widgets to your blog/site. Tweet and retweet quality stuff..

I got 703 followers in the past week. The way I do it is this

1 I follow as many people as Twitter will let me. I think I can follow 10% more than I started with.
2 I leave it for 2 days and in that time follow all new followers who follow me. I follow them back from following the Reciprocate button on Tweepi
3 I unfollow anyone who is inactive or who hasn't followed me back using manageflitter.com. I unfollow these people two days after I have added them.
4 Repeat the process again by following as many people as Twitter lets me (another 10% on top of the people I am already following).

Managing your following by adding new followers and cutting back on unfollowers and inactive followers is essential. Think about it like a tree. You need to manage it by letting it grow and pruning it when you need to - just like that.

I now have over 6000 real followers.

It is always good to retweet quality tweets and favorite all tweets that you want to return to later. Spend some time reading the different tweets of the people you follow and interacting with those you find interesting.

Some great advice here, but here is one that has not explicitely been pointed out. There are many twitter chat groups out there (like #foodiechats) and it seems almost one for every niche. Participate in these chats and it really helps you build relationships with likeminded individuals... once engaged, many will follow you.
