As far as SEO is concern which design is best now days?

CSS, html, or xhtml ?

Does it make a difference?

    you can use anyone , but make sure that that there is no flash and website is easy to load.

    Also make sure that whatever way you are using your site passes the w3c validator, it is supposedly helps with SERPs, but I guess it is a minor factor.

    All of these are fine for SEO. You have to make sure that your website loads quickly. Avoid using Frames and flash. Upgrade your website with latest technology updates. Eg. HTML5

    HTML and XHTML are markup languages and you add the "design" via CSS.
