How to improve my website conversion rate?

I'm thinking of redesigning my website main page to improve the sign up rate. But I have little idea on how and what changes I need to make.
Questions
What should be in the main page?
Should I add a landing/splash page?
Will adding flash intro improve the sign up?
I designed this myself but the more I see it the more I dont like it...I'm sure its not giving the visitors the best impression and any clues what is the website all about.
