    It is a forum or blog?

    For forum, you can directly moderate the new users and if it is a blog, you can change the settings of comments in comments section.

    If you are using a CMs, there are some modules/plugins for preventing spams. For example Wordpress, has a good plugin which is called Akismet that prevent spams well. So you should find it according to the CMS you are using on your site. There is not a general plugin for all platforms and each of them have someones.

    I have always used Akismet and it does a good enough job, but I just converted a couple of old HTML sites to WordPress and I am trying a new plugin called Growmap Anti Spambot Plugin. It ads a client side checkbox to the comment form requesting that the user clicks it to prove they are not a spammer. It eliminates spam bots from auto commenting. It works very well.
