1) Optimize your video for relevant keywords (video file, title, description, tags), of course. This will take care of search.

2) Take time to find out where you want your video to show up in "Related Videos." That is, assess what videos in your niche are currently getting traffic and analyze the keywords they use. Match some of those keywords and try to create an obvious connection between those videos and your videos, so YouTube places them in the "Related" category.
