I want make a file hosting site like mediafire. Can I do it with a regular domain and hosting? If yes, is there any new method for creating it?

    I'd be particularly mindful of the hurdles involved in hosting and running a file-sharing website. You, as the owner, are going to be receiving (potentially) many DMCA take-down notices from copyright holders. You will need to carefully monitor all files to ensure that no one uploads illegal content, because if you do not act promptly, you could end up like Dotcom did (from MegaUpload). Although, his case is pretty unique (major game-player, apparently ignored DMCA requests, etc). Running a file-hosting website is a very risky and daunting task, especially with the eyes of the Government and their best friends the MPAA/RIAA looking around.

    I doubt any hosting provider will have obligations to you hosting your own content, however if they begin receiving any DMCA take-down notices or copyright infringement complaints, they may drop you. I'd suggest finding a hosting provider out of your own country (especially the US).

    In most cases no, you cannot. I'd suggest doing a little research as you will most likely come across a lot of legal issues with content & materials that you're hosting. You'll also need billing software, servers, storage etc.

      Actually can't we dodge this legal and illegal issue with content & materials cause I will only accept game files in my hosting site.
